Abstract

The objective of this study was to determine the effects of treatment on the effect of sildenafil citrate (Viagra) on the erectile response of male rats with erectile dysfunction. Male Sprague-Dawley rats were treated orally with sildenafil citrate (Viagra) or vehicle for three days (3 and 12 h). After the third day of treatment, the erectile response to the drug was studied. Male Sprague-Dawley rats treated with sildenafil citrate for up to 12 h (3 and 12 h) had a significantly lower incidence of adverse events (ejaculatory latency time (EAT)) and a significantly longer duration of erection than those treated with vehicle (0 and 1 mg/kg, respectively). Sildenafil citrate treatment significantly improved the incidence of adverse events (EAT) in male Sprague-Dawley rats treated with sildenafil citrate (3 and 12 h) compared to the vehicle-treated rats (3 and 12 h). In addition, sildenafil citrate treatment significantly reduced the duration of erection of the rats by up to 30% (p < 0.001) compared to the vehicle-treated rats. However, the treatment with sildenafil citrate significantly improved the duration of erection in the rats with erectile dysfunction.

Background:Sildenafil citrate has been shown to be effective in treating erectile dysfunction by enhancing the phosphodiesterase type 5 (PDE5) enzyme. Erectile dysfunction is the inability to achieve or maintain an erection sufficient for sexual intercourse. PDE5 is a key enzyme that regulates blood flow and is activated by prostaglandins. After the third day of treatment, the EAT of the treated rats was determined. After the third day of treatment, the rats were sacrificed. Male Sprague-Dawley rats were either treated orally with sildenafil citrate (Viagra) or vehicle for up to three days (3 and 12 h). Male Sprague-Dawley rats were pretreated with vehicle or sildenafil citrate (Viagra) for three days (3 and 12 h). The rats were then used to perform the experiment.

Results:In the rats treated with sildenafil citrate (Viagra) for up to three days, sildenafil citrate significantly reduced the incidence of adverse events (EAT) in the rat test model. In addition, sildenafil citrate significantly improved the incidence of adverse events in the rats with erectile dysfunction.

Conclusions:The effects of sildenafil citrate on the EAT of male Sprague-Dawley rats with erectile dysfunction were dose-dependently improved. The treatment of sildenafil citrate (Viagra) increased the PDE5 enzyme activity, which was associated with the inhibition of the phosphodiesterase type 5 (PDE5) enzyme. These results support the concept that sildenafil citrate may exert its action on the PDE5 enzyme.
